TERA has
developed two documents addressing broad issues related to children’s risk,
which will be discussed at upcoming peer consultation meetings.
One
document presents a draft
framework for evaluating the adequacy of the default uncertainty factor of
3.2 for human kinetic variability for protecting children, and the
second one addresses uncertainty factor selection when deriving safe dose
estimates to address reproductive or developmental toxicity concerns.
TERA has published on
age-related differences in toxicokinetics and chemical sensitivity, and how
these issues are taken into account in the current risk assessment paradigm (Dourson
et
al., 2002;
Scheuplein
et al., 2002). To help
address the key issue of how tissue dose to children and adults compares for a
given environmental exposure, TERA has collected physiological parameters for neonatal animals for
use in PBPK modeling (Gentry
et al., 2005). These data
can be used to develop age-specific models to improve estimation of risk to
children.
TERA is also organizing peer consultations for the
Voluntary Children’s Chemical Evaluation Program (VCCEP).
This work provides ongoing contact with a broad range of leading
scientists and their perspectives on addressing children’s risk from the
hazard identification, dose-response and exposure perspectives.
